Detection and identification of Candidatus Liberibacter asiaticus in Citrus limon L.

Mexico has a production of more than 8 million 217 thousand tons of citrus fruits distributed over 600 thousand hectares. The main citrus fruits produced are: orange, Mexican limon, Persian lime, and mandarin. Citrus fruits are threatened by the presence of the bacteria Candidatus Liberibacter asiaticus, which causes the disease known as Huanglongbing (HLB), or yellow dragon, or yellow shoot disease. For this reason, the objective was to determine the number of bacterial cells of Candidatus Liberibacter asiaticus in Citrus limon. 53 Citrus limon plants grafted onto Citrus aurantium L., Citrus volkameriana, and Citrus macrophylla rootstocks were used. The stem was inoculated with the bacteria Candidatus Liberibacter asiaticus; after six months, DNA was extracted from the central veins and leaf petioles. Among 18 DNA samples extracted from Citrus limon on Citrus volkameriana rootstock, 14 were negative for the pathogen Candidatus Liberibacter asiaticus and 4 were positive for it, in addition 20 DNA samples from Citrus limon on Citrus aurantium rootstock, 6 samples were positive and 14 were negative for the pathogen, likewise 16 DNA samples extracted from Citrus limon on Citrus macrophylla rootstock, 11 were negative for the pathogen Candidatus Liberibacter asiaticus and 5 were positive for it.
